No, you can spend Crystals during different Moves. As long as the total number of Crystal spent during your Rival’s Moves this Round are greater than or equal to 2, you can use this effect.
What does "Double Defended" mean?
It means a card must be defended by 2 Allies who match their attack color(s) or 2 Allies who don’t.
